A New Technique for the Extraction of Arbuscular Mycorrhizae Fungal Spores from Rhizosphere

Monitoring the dynamics of spore bank arbuscular mycorrhizal fungi (AMF) is essential for sustainable management and protection agroecosystems. The most common method extracting AMF spores from soil wet-sieving technique (WST). However, this has many disadvantages. In study, we modified WST using new approaches: ultrasound (UWST) centrifuge (UCT). We enumerated compared numbers quality obtained WST, UWST, UCT to validate techniques. extracted rhizospheres different plants, including wheat (Triticum aestivum L.), bean (Phaseolus vulgaris tomato (Solanum lycopersicum pepper (Piper nigrum parsley (Petroselinum crispum Mill.), turfgrass (Lolium perenne L.) collected Van Lake basin, Turkey. highest lowest were observed in rhizospheres. allowed extraction number all rhizospheres, followed by UWST WST. similar wheat, pepper, parsley, Beyond high number, was shown be a fast low-material-consuming approach.
